Glibc is automatically added as an input, under the name “glibc” (see
build-system/gnu.scm).
So you can just do something like:
(lambda* (#:key inputs #:allow-other-keys)
(substitute* "setup.py"
(("/usr/include")
(string-append (assoc-ref inputs "glibc") "/include"))))
