Like this:
  $ cd ~/emacs/build
  $ rm -fr *
  $ CPPFLAGS='-DGLYPH_DEBUG=1' CFLAGS='-O0 -g3'
../emacs.git/nt/msysconfig.sh --enable-checking

And apparently, everything was fine:
