I assume you mean the file is *in* the 'C:\' directory and is called '.emacs'
(and is not in a directory called 'c:\.emacs').

Some checks you can do:

1. Check if you have defined a HOME variable that points to a location other
   than C:\. This location will override C:\ as the default location for
   checking the .emacs file.

2. See if the file permissions (or 'attributes') are correct for .emacs. You can
   check this through the properties menu item in the context menu from
   Explorer. Especially check if your user ID has the right access.
