Yours would have to be more like
(defun isupper (char)
(let (case-fold-search)
(string-match-p "[[:upper:]]" (make-string 1 char))))
but has the advantadge that
ELISP> (isupper ??)
nil
ELISP> (eq (upcase ??) ??)
